5、摘要目的食管鳞癌是人类十大恶性肿瘤之一,其死亡率居我国恶性肿瘤死因第四位。失巢凋亡是内皮及上皮细胞脱离细胞外基质发生细胞凋亡的一种特殊的死亡形式,细胞获得抵抗失巢凋亡能力是肿瘤细胞发生远处转移的前提条件之一。虽然一些研究提示,Maspin、SKP2、xCT、PTTG1、TM4SF3、ECRG2、CTTN和CyclinB1等基因表达的改变可影响ESCC细胞在体内的致瘤及转移能力,本课题组前期在食管磷癌研究中也已发现多个抗失巢凋亡相关的癌基因,尽管这些基因都可参与食管鳞癌细胞失巢凋亡抗性的调节,但它们是通过不同的信号通路发挥作用,这些结果显示食管鳞癌细胞中存在复杂
6、的信号网络以适应宿主微环境,从而赋予食管鳞癌细胞更强的转移潜能。本实验主要目的是在食管磷癌中寻找更多与抗失巢凋亡相关的基因,以进一步阐明食管鳞癌细胞转移的分子机制,开发有效的诱导肿瘤细胞失巢凋亡的策略,从播散环节阻止肿瘤发生远处转移,有可能成为预防和控制肿瘤扩散的一条有效途径。方法采用逆转录病毒文库功能筛选的手段,首先构建食管磷癌细胞系的逆转录病毒文库,并感染对失巢凋亡较敏感的NIH3T3细胞,利用感染病毒cDNA文库中混合细胞系进行软琼脂集落形成实验,挑取出在悬浮条件下仍可生长形成较大集落的细胞单克隆(即潜在具有抵抗失巢凋亡能力的细胞),最后通过逆转录病毒载
7、体特异引物PCR扩增失巢凋亡抗性克隆基因组中的外源插入片段,在食管癌细胞系cDNA文库中获得潜在的具有失巢凋亡抗性的基因。结果发现经食管癌逆转录病毒cDNA文库感染的NIH3T3细胞经软琼脂悬浮培养后,大部分克隆形成的多个抗失巢凋亡集落与感染pMSCV-GFP的NIH3T3细胞形态有明显差别,挑取直径明显大于对照组的克隆细胞,将获得的失巢凋亡抗性克隆扩大培养,提取基因组DNA,采用逆转录病毒特异性的pMSCV5’和pMSCV3’引物,扩增靶细胞基因组中的外源插入片段,结果显示亲本NIH3T3细胞组未扩增出产物,感染pMSCV-cDNA的靶细胞组扩增了食管-2-
