1、重庆医科大学硕士学位论文高脂饲料诱导肥胖大鼠网膜素水平及外源性网膜素的干预作用姓名:吕攀攀申请学位级别:硕士专业:儿科学指导教师:刘友学201205重庆医科大学硕士研究生学位论文高脂饲料诱导肥胖大鼠网膜素水平及外源性网膜素的干预作用摘要目的:利用高脂饲料喂养大鼠,建立肥胖大鼠模型,比较大鼠在高脂饲料喂养下的体重变化情况、血生化特点、血清网膜素水平及网膜脂肪组织网膜素mRNA表达水平;观察注射外源性网膜素后,肥胖大鼠上述指标的变化,同时探讨外源性网膜素是否对肥胖大鼠具有干预作用。材料与方法:40只SD健康雄性大鼠随机分组:普通饲料组(正常组,n=
2、lO),另外30只采用高脂饲料喂养,每周检测体重,高脂饲料喂养6周后选取体重增长值最大的20只,随机分为两组,对照组(n=10)和干预组(n=10)。干预组给予尾静脉注射外源性网膜素O.1ml(6腭),正常组及对照组尾静脉相同体积的注射生理盐水。正常组继续普通饲料喂养,对照组和干预组继续高脂饲料喂养,喂养至10周后再次按上述方案注射,干预组尾静脉注射外源性网膜素剂量为3一只,继续喂养至12周。实验结束时三组各剩9只,检测体重后从心脏取血测定空腹血糖(Glucose,GLU)、血清总胆固醇(Totalcholesterol,TC)、甘油三酯(Tr
3、iglyceride,TG)、高密度脂蛋白(High.densitylipoprotein,HDL)、低密度脂蛋白(Low-densitylipoprotein,LDL)、空腹胰岛素(Insulin,INS)、血清网膜素。同时取网膜脂肪组织检测网膜素mRNA4重庆医科大学硕士研究生学位论文表达水平。取颈总动脉做病理切片检测动脉内膜厚度。结果:(1)喂养12周后,高脂饲料喂养大鼠(对照组)的体重、体重增加值及肥胖指数(Obesityindex,OI)均高于正常饲料喂养组(分别为591.71VS525.98g,286.59VS211.899,0.0
