3、方法:体外培养人脐静脉内皮细胞株,选第3—9代细胞用于实验。将人脐静脉内皮细胞与加载盐负荷的RPMI1640培养基(氯化钠终浓度提高30mmol/L)共同孵育不同时间(6h、12h、24h、48h)作为测定组;将与测定组等体积的D-Hank’S液代替NaCl加入细胞培养液中,培养细胞48h,作为空白对照组;应用等毫摩尔甘露醇代替氯化钠培养细胞48h,并检测各项指标以排除实验干扰因素。细胞爬片并进行HE染色,于显微镜下观察细胞形态变化;CCK-8试剂比色法检测细胞的生长活性;酶标仪检测乳酸脱氢酶漏出量;采用一氧化氮试剂
4、盒(硝酸还原酶法)比色法检测细胞培养液中一氧化氮的含量;生化法检测NOS活性;细胞爬片,免疫细胞化学技术观察eNOS和iNOS在蛋白水平的表达;采用逆转录-聚合酶链反应技术观察eNOS和iNOS在基因水平的表达。实验数据采用均数±标准差(x±s)表示,使用SPSS19.0统计软件进行统计学分析。结果:盐负荷能导致人脐静脉内皮细胞发生形态学改变和凋亡,细胞的生长活性受到抑制,细胞LDH漏出量增加,且呈时间效应关系;盐负荷呈时间依赖性地诱导内皮细胞培养液中一氧化氮的含量增加,于24h达高峰;盐负荷能抑制内皮型一氧化氮合
5、酶的活性,及其在蛋白水平和基因水平的表达,且呈时间依赖性;盐负荷呈时间依赖性地引发诱导型一氧化氮合酶的活性增1强,及其在蛋白水平和基因水平的表达增加,于24h时达高峰。通过应用等毫摩尔甘露醇代替氯化钠培养细胞并对各项指标进行检测,证明一氧化氮、LDH、eNOS和iNOS的改变不是由于加载盐负荷后渗透压改变而引起的。结论:1、盐负荷能直接诱导人脐静脉内皮细胞损伤。2、盐负荷诱导人脐静脉内皮细胞发生损伤的机制可能是:eNOS的活性及表达受到抑制,生理性一氧化氮的合成减少,内皮细胞出现损伤;同时,盐负荷诱导iNOS的活性及
