1、提要本文对桑籽降血糖有效物质的提取、纯化、药理活性的验证等环节进行了系统试验研究,确定了合适的有效物质提取纯化方法,验证了其药理活性。黄酮、生物碱和多糖是桑资源中的是比较重要的三类活性成分。本文建立了桑籽中脂肪油、总生物碱(DNJ)、总黄酮、总多糖快速、简便的含量测定方法,对不同的提取方法进行了比较,确定了合适的提取方法;在此基础上,通过大孔树脂-聚酰胺树脂联用,确定了对总黄酮具有良好吸附的树脂型号及其分离富集工艺;又对总多糖进行醇沉、除蛋白、透析,确定了总多糖的富集工艺;然后对生物碱进行除蛋白、50%醇沉除杂、正丁醇萃取,确定了生物碱的富集工艺,最后通过单个有效部位及有效部位之间的
2、配伍确定最有效的降血糖组份。实验结果表明,桑籽中的部位经已提取分离工艺提取后,其总黄酮纯度约为52.3%,总多糖纯度为64.4%,总生物碱纯度为35.6%,经药理活性验证,生物碱和多糖具有良好的降血糖活性,为桑籽胶囊的研制和桑保健食品的开发提供了试验依据。关键词桑籽;提取;纯化;药理活性;工艺研究ResearchonExtractionandPurificationandpreIiminarypharmacologicalActivityofhypoglycemicSubstancefromMulberrySeedSpeciaIity:PharmacyofTraditionalChi
