1、AbstractPlantsbelongingtothegenusLabiataeIsodonarewidelydistributedinChina.Therearevarietyofcompounds,whichisisolatedfromLabiataeIsodon,applytoanti-tumortherapybasedontheirhighefficiencyandlowtoxicity.Ent-kaurenediterpenoids,whicharethemainactiveconstituentsagainsttumor,

2、arerichinthegnusIsodon.Mostofthemareusedasantitumor,antibacterial,andanti-inflammatoryherbs.Inrecentyears,itwasreprtedthatOridonin,akindofEnt-kaurenediterpenoids,havegoodcurativeeffectontreatmentofleukemia.However,thecompoundswhichhavesimilarmolecularstructureofEnt-kaure

3、nediterpenoidsagainstleukemiahaverarelyreported.Manyofthemexploretheantitumormechanismthroughtheapoptoticpathway.LeukameninEisseparatedfrom(Isodonexcisoides(SunexC.H.Hu)C.Y_WuetH.w.Li)inGansuProvince,whichisoneofent-kauranediterpenoids,thereisresearchshowsthatthiscompoun

4、dhasstrongcytotoxicity.Buttheantitumormechanismisalsonotclear.Atpresent,therearefewliteraturereporttheeffectofLeukameninEonthecytoskeleton.LeukameninE,anent-kauranediterpenoid,isusedassubjectscompoundtotesttheeffectsonHL-60cellgrowthwithtrypanblueexclusionmethodandMTT.Th

5、eeffectofLeukameninEonproliferationandapoptosisinductionwerestudiedbyFCMandAO/EB.Additionally,Hoechst33258fluorescentstainingandchromosomeaberrationtestwereusedtoevaluatetheproductionofnucleusandchromosomeaberrationinHL-60cells,respectively.Atlast,weobservethechangeofcyt

6、oskeletonwithdifferentconcentrationofLeukameninE.Theresultsasasfollows:1.LeukameninEhasstrongercytotoxicityonHL-60cells,restrainscellgrowthunderlowconcentration,andhaslethaleffectsunderhighconcentrationandatime-anddose-dependentmanners.MTTassayshowthatIC50=0.89±0.15µmol/

7、L.2.PIstainingassaysbyflowcytometrydisplayedthatthedistributionofcellcyclecouldbechangedwiththetreatmentofLeukameninE.ItcouldinducestrongSphasearrestintime-dependent.ThefragmentofDNAcouldnotoverflow.Atlast,thecellsarrestSphaseuntildead.SoLeukameninEcouldinducetheapoptosi
