5、xtractedbythemethodsofwateextracting.FactorsthathadinfluenceontheyieldofPRPMweremainlyconsidered,includingextractiontemperature,extractiontime,extractiontimes,thesolvent-solidratio,etc.Onthebasisofsinglefactorexperiments,orthogonaldesignwasadoptedtosearchtheop
6、timumextractingparameterofPRPM..Theresultshowedthatoptimumextractingparameterasfollows:90℃ofextractiontemperature,1hofextractiontime,2timesofextractiontimes,15:1ofthesolvent-solidratioandpH7.0weresuitable.Takingglucoseascontrol,thephenol-sulfuricacidmethodexam
7、iningcontentofPRPMwasestablished.Theresultsshowedthattherevisedmethodwasstableandhadgoodrepeatabilityandhighaccuracywhenthevolumeratioof5%phenoltosulfuricacidwas1:4,thevolumeratioofsulfuricacidtototalvolumeofsolutionwas2:3andreactionsolutionwascooledafterbathi
8、ng15mininboilingwater.TheaveragecontentofPRPMwas18.54%.Thesecondpart:TwomethodsofsevegandTrichloroaceticacid-N-ButanolareusedtoremoveproteinfromPRPM.Theresultsshowthatthemethodofse
