6、······························································43个人简历··········································································································44中文摘要七氟醚诱导老龄大鼠认知功能障碍时自噬与凋亡关系摘要目的:本研究拟探讨七氟醚诱导老龄大鼠认知功能障碍时自噬与凋亡的关系。方法:选取健康雄性SD大鼠120只,18月龄,体重500~550g,采用随机数字表法将其分为4组(n=30

7、):对照组(C组),认知功能障碍组(P组),自噬抑制剂3-甲基腺苷组(MA组)和自噬激动剂雷帕霉素组(R组)。MA组大鼠尾静脉注射自噬抑制剂3-甲基腺苷(3-MA)1mg/kg,R组大鼠尾静脉注射自噬激动剂雷帕霉素2mg/kg,C和P组大鼠尾静脉注射等量的生理盐水。给药结束30min后,P、MA和R组大鼠暴露于3%七氟醚3h,C组大鼠不做任何处理。于麻醉前1d、麻醉后3和7d行Morris水迷宫实验检测大鼠认知功能。每个时间点水迷宫实验结束后取大鼠海马组织,采用流式细胞术检测海马神经元凋亡率
