1、大鼠肾虚型颈椎病模型的建立【摘要】采用病、证模型复合的方法建立大鼠肾亏型颈椎病动物模型。方法:选择3月龄雌性SPF级SD大鼠30只,随机分为正常组、颈椎病模型组和肾虚型颈椎病模型组,每组10只。采用动静力失衡性大鼠颈椎间盘退变模型复合去卵巢肾虚亏模型的方法建立肾虚型颈椎病模型。通过检测大鼠子宫及附件形态和质量、血清雌二醇(estradiol,E2)含量验证肾虚证;通过颈椎间盘组织病理学、Ⅱ型和Ⅹ型胶原的免疫组化检测及聚集蛋白聚糖(aggrecan1,Agc1)、Ⅱ型前胶原基因(typeⅡprocollage
2、ngene,Col2a1)、基质金属蛋白酶13(matrixmetalloproteinase13,MMP13)和基质金属蛋白酶抑制剂1(tissueinhibitorofmetalloproteinases1,TIMP1)的基因表达情况评判椎间盘退变。结果:与正常组和颈椎病模型组相比,肾虚型颈椎病模型组动物子宫及附件形态萎缩、质量减轻,血清雌二醇含量降低,颈椎间盘组织病理学退变更加明显,Ⅱ型胶原蛋白表达减少,Ⅹ型胶原表达增高,Agc1和Col2a1基因表达降低,MMP13基因表达增高。结论:通
