1、GDNF在慢性收缩性损伤大鼠脊髓背根神经节中的蛋白表达作者:夏小萍,马正良,朱巍,周锦勇【摘要】目的探讨胶质细胞源性神经营养因子(GDNF)在慢性收缩性损伤(CCI)所致神经病理性疼痛大鼠的L4背根神经节中的蛋白表达。方法实验1:24只SD大鼠,随机分为正常组(Naive组)、假手术组(Sham组)和CCI组(每组n=8)。Naive组不做任何手术;Sham组仅暴露坐骨神经;CCI组按Bennett法结扎右侧坐骨神经主干制备CCI模型。分别测定Sham组和CCI组术前2天(基础值)及术后第1、3、5、7、14、21天(Naive组
2、则在相对应的时间点)大鼠同侧(本实验为右侧)后足机械缩足反射阈值(MWT)和热缩足反射潜伏期(PWL)。实验2:18只SD大鼠随机分为正常组(Naive组)、假手术组(Sham组)和CCI组(每组n=6)。CCI组制备CCI模型,术后第7天处死大鼠,取同侧L4背根神经节;Naive组不做任何手术,直接取右侧L4背根神经节;Sham组仅暴露坐骨神经,术后第7天处死大鼠,直接取右侧L4背根神经节。采用Westernblot法检测脊髓L4背根神经节GDNF的蛋白表达。结果实验1:与Sham组相比,CCI组大鼠从术后第3天开始至术后21天
