2、athology,El-MiniaFacultyofMedicine.ABSTRACT:Background:Breastcancerisamultistepprocess.PreviousstudiesshowedthatelevatedROCK-1activityleadstoincreasecancercellsinvasionandmetastasis.Reducedexpressionofp27associatedwithcancerprogression.Aim:TostudytheimmunohistochemicalexpressionsofROCK-
3、1,p27,ERandPRinapanelofproliferativeepitheliallesionsoffemalebreast.Materialsandmethods:Thecurrentstudyincludesatotalof100randomlyselectedproliferativeepitheliallesionsoffemalebreast.Formalinfixedandparaffin-embeddedtissuesectionsfromcasesunderinvestigationweresubjectedtohaematoxylinand
4、eosinstaining,andimmunohistochemicalstainingforROCK-1,p27,ERandPR.Results:ProteinexpressionofROCK-1wasdetectedin50.6%ofmalignantinvasivelesions,25%ofDCISandin6.7%ofbenignproliferativelesions.Itwassignificantlyassociatedwithsomevariablesofmalignantinvasivelesionsincludinghistologicaltype
5、s,NPI,lymphnodestatus,ERnegative,PRnegativeandcombinedER-/PR-immunophenotype.Reducednuclearp27immunostainingwasdetectedin54.5%ofmalignantinvasivelesions,in25%ofDCISandin6.7%ofbenignproliferativelesions.Itwassignificantlyassociatedwithsomevariablesofmalignantinvasivelesionsincludinghisto
6、logicalsubtypes,histologicalgrade,NPI,pathologicalsize,lymphnodestatus,stages,ERnegative,PRnegativeandcombinedER-/PR-immunophenotype.Cytoplasmicp27immunostainingwasdetectedin61%ofmalignantinvasivelesions,in50%ofDCIS;whilenocaseofbenignproliferativelesionsshowcytoplasmicp27immunostaining
7、.Itwassignificantlyassociatedwiththefollowingvariablesofmalignantinvasivelesions,histologicalsubtypes,histologicalgrade,lymphnodestatus,ERstatus,PRstatusandcombinedER/PRimmunophenotype.AnegativecorrelationwasfoundbetweenROCK-1andnuclearp27immunostainingindifferentstudycategorie
