1、第27卷第6期长治医学院学报Vo1.27NO.62013拒12月JOURNALOFCHANGZHIMEDICALCOLLEGEDec.2013417粗叶悬钩子总生物碱对非酒精性脂肪肝大鼠脂代谢及CPT工的影响伍娟钟晓勇赵锦燕郑玉清郑海音洪振丰摘要目的:探讨粗叶悬钩子总生物碱(TARAP)对非酒精性脂肪肝(NAFLD)大鼠脂代谢及肝脏形态学的影响,研究其作用机制。方法:购入的SD大鼠适应性喂养7d,然后按照体重分层随机分为正常组、模型组、东宝肝泰组、TARAP低剂量组和高剂量组,正常组饲以基础饲料,饮用自来水,其余各组饲以高脂饲料,持续喂养12周。造模成功后各组予以相应药
2、物灌胃4周后取材,HE染色,观察肝组织病理形态学变化。RT—PCR和免疫组化法检测肉碱脂酰转移酶I(carnitinepalmityltransferase工,CPTI)的基因和蛋白表达。结果:TARAP低剂量组、高剂量组脂肪变细胞较模型组明显减少;RT-PCR和免疫组化结果显示,TARAP能显著提高CPTI的表达。结论:TARAP能提高CPTI的表达,减轻肝细胞脂肪变性,提示TARAP通过调节CPTI的表达是其治疗NAFLD作用机制之一。关键词粗叶悬钩子总生物碱;非酒精性脂肪肝;CPTI中图分类号R285.5文献标志码A文献编号1006一(2013)06—417—04
