1、高迁移率族蛋白N2对人舌癌裸鼠移植瘤抑瘤作用探究 [摘要]目的研究高迁移率族蛋白N2(HMGN2)对人舌癌裸鼠移植瘤生长的抑瘤作用。方法裸鼠腋下注射接种舌鳞癌细胞Tca8113以建立裸鼠移植瘤模型,接种7d成瘤后,设立阴性对照组、HMGN2蛋白组和阳性对照组,在瘤体周边分别注射含washingbufferⅡ、HMGN2蛋白和顺铂的细胞培养基。观察各组裸鼠肿瘤的生长情况,经过4次给药后处死裸鼠,取出瘤块,称其质量计算抑瘤率,并行苏木精-伊红(HE)染色观察形态学变化。结果成功建立舌癌裸鼠移植瘤模型。HMGN2蛋白组和阳性对照组肿
2、瘤体积明显小于阴性对照组。裸鼠体重在整个实验过程中并没有明显的变化。阴性对照组、HMGN2蛋白组、阳性对照组的肿瘤质量平均值分别为(0.38±0.19)、(0.21±0.15)、(0.23±0.16)g,3组间的差异无统计学意义。HMGN2蛋白组和阳性对照组的抑瘤率分别为45.71%和39.44%。肿瘤经肉眼观察可见阴性对照组瘤块较大,HE染色可见HMGN2蛋白组和阳性对照组细胞坏死明显。结论HMGN2蛋白可以明显抑制人舌癌裸鼠移植瘤的生长。[关键词]高迁移率族蛋白N2;移植瘤;Tca8113细胞10[中图分类号]R730.2[
