2、验第0、7、14天分别以粉尘螨提取液腹腔致敏,共3次,对照组以PBS代替。于实验第28天起,哮喘组小鼠以粉尘螨提取液滴鼻激发,连续7天,观察并记录小鼠哮喘发作情况;对照组以PBS代替。最后1次滴鼻24h后对小鼠作肺泡灌洗液。进行灌洗液细胞计数和分类,肺组织病理学检测(HE染色),ELISA测定血清总IgE和重组粉尘螨二类变应原特异性IgE(rDerf2-sIgE)含量,ELISA试剂盒测定支气管肺泡灌洗液(BronchoalveolarLavageFluid,BALF)中IL_4和IFN一的含量。结果:哮喘组小鼠肺泡灌洗液中细胞总数和嗜酸性粒细胞较正常组极显著增
3、加(P<0.01);肺组织可见明显的炎性细胞浸润;血清总IgE(P<0.001)和rDerf2-sIgE(P<0.01)水平极显著升高;BALF中IL-4水平显著增高(P<0.05),而IFN一的水平极显著下降(P<0.01)。结论:使用多次腹腔致敏和滴鼻激发的方法可以建立产生高水平IgE的粉尘螨过敏性哮喘动物模型[关键词]IgE;粉尘螨;哮喘;动物模型PreparationandevaluationofahighlevelofIgEofmouseasthmamodelinducedbyDermatohpagoidesfarinaeextractsRANWei,
