1、中国神经精神疾病杂志2014年第40卷第12期741论著·甲强龙预处理对大鼠脊髓损伤后claudin一5表达的影响☆崔敏周昌龙贺学农夏小辉张光伟【摘要】目的探讨甲强龙(118,17旺,21一三羟基一60L一甲基孕甾一1,4一二烯一3,20二酮一21一琥珀酸钠)对大鼠脊髓损伤(spinalcordinjury,SCI)后血一脊髓屏障(bloodspinalcordbarrier,BSCB)通透性及紧密连接蛋白claudin一5表达的影响。方法参照改良Allen’S法建立大鼠脊髓损伤模型。分为假手术组(n=5
2、0)、脊髓损伤组(n=-50)、甲强龙预处理组(n=50)。建模后不同时间点(12h、ld、3d、5d、7d)各组随机取5只大鼠检测BSCB通透性,取胸段脊髓损伤区组织经RT—PCR、Westernblot方法检测claudin一5表达。结果本实验制模成功率84.O%;脊髓损伤组各时间点EB含量较假手术组显著增高(F=27.732,P均<0.05)。甲强龙预处理组各时间点EB含量较脊髓损伤组显著降低(F=48.149,P均<0.05);脊髓损伤组各时间点claudin一5mRNA表达较假手术组显著降低(1
