1、2014年5月第4卷第9期·论著·复方守宫散对Lewis肺癌小鼠JAK2-STAT3信号通路的影响1△2▲211沈迪李崇慧周晋华徐海惠周鹏飞1.安徽中医药大学,安徽合肥230000;2.安徽中医药大学第一附属医院肿瘤科,安徽合肥230000[摘要]目的观察复方守宫散对Lewis肺癌小鼠JAK2-STAT3信号通路的影响。方法将55只C57BL/6J小鼠按常规方法接种Lewis肺癌瘤株,6d后选取荷瘤成功的小鼠50只随机分为5组:阳性对照组、西药组、中药高剂量组、中药中剂量组、中药低剂量组,每组10只。阳性对照组:灌服生理盐水0.2mL,1次/d;中药组灌服复方守宫散高剂量组0.6mL
2、(相当于120mg/kg),中剂量组0.4mL(相当于60mg/kg),低剂量组0.2mL,(相当于40g/kg),1次/d;顺铂组按1mg/kg给药,0.1mL/只腹腔注射。每天1次,共14d。在光镜下用HE染色法观察肿瘤组织病理形态学改变情况,用WesternBlot法检测JAK2、STAT3蛋白的表达水平。结果各给药组JAK2、STAT3蛋白的表达水平出现不同程度降低,且JAK2降低较为明显。结论复方守宫散能够抑制肿瘤生长,降低Lewis瘤细胞中JAK2、STAT3蛋白的表达,并通过此方式阻断JAK2-STAT3信号转导。[关键词]肺癌;信号通路;实验研究[中图分类号]R734
