1、10TianinMedJ，Jan2014，Vol42Nol土槿乙酸对人肝癌细胞中血管内皮生长因子表达的影响祝平郭莲怡【摘要】目的探讨土槿乙酸(PAB)对人肝癌细胞BEL-7402中血管内皮生长因子(VEGF)表达的影响。方法以O(对照组)、0．5、1．0、2．0、4．0、8．0I~mol／LPAB处理人肝癌细胞BEL一7402，培养6h，0．4％台盼蓝染色，在显微镜下观察并计算细胞活率。采用WST一8法检测细胞增殖，RT—PCR法检测人肝癌细胞BEL一7402中VEGFmRNA表达，WesternBlot检测VEGF蛋白表达。结果不同剂量PAB处理的人肝癌细胞BEL一7402的细

2、胞增殖率与对照组比较差异无统计学意义。随着PAB浓度的增加，VEGFmRNA表达与对照组(2．53±0．07)相比分别降低至1．90+0．02、1-31±0．03、0．86~0．04、0．60~0．03、0．34+0．02，差异有统计学意义(P<0．O1)，VEGF蛋白表达与对照组(3．06~0．15)相比分别降低至2．60~0．09、2．42_+0．07、1．85~0．05、0．92~0．05、0．60+0．04，差异有统计学意义(P<0．01)。结论PAB可抑制人肝癌细胞BEL一7402中VEGFmRNA和蛋白的表达。【关键词】细胞系，肿瘤；肝肿瘤；血管内皮生长因子类；逆转录
