1、中华实验眼科杂志2014年9月第32卷第9期ChinJExpOphthalmol,September2014,Vo1.32,No.9·实验研究·去卵巢大鼠结膜组织中炎性因子的表达变化及干眼相关检查的评估钟蕾张晓峰夏静孙正太王英明夏蔚【摘要】背景近年来,干服的发生率逐年增加,其病因及眼部表现多样,研究认为女性绝经是干眼发病的高危因素之一。目前关于炎性因子参与干眼发病的病理机制越来越受到重视,探讨炎性因子,如白细胞介素(IL)和肿瘤坏死因子(TNF),与性激素改变的关系对干眼病因学和病理机制的探讨具有重要意义。目的建立去卵巢大鼠模型,观
2、察性激素水平变化对结膜炎性因子表达的影响。方法将健康成年雌性SD大鼠2O只按随机数字表法随机分为卵巢摘除组和伪手术组。卵巢摘除组大鼠行双侧卵巢摘除术,伪手术组大鼠不摘除卵巢,其他手术步骤同卵巢摘除组。两组大鼠均于术前及术后1、2、3个月行Schirmer试验I(sIt)和角膜荧光素钠染色检查,评估眼表干眼症状。于术后3个月时大鼠行心脏穿刺采血,用放射免疫分析法检测两组大鼠血清雌二醇、睾酮的质量浓度。以过量麻醉法处死大鼠,收集大鼠结膜组织行苏木精一伊红染色,观察大鼠结膜上皮细胞的形态学变化,分别采用免疫组织化学法和Westernblo
3、t法检测两组大鼠结膜组织中IL.17A、IL一1B、IL.6、TNF.d蛋白的表达。结果卵巢摘除组和伪手术组大鼠术后3个月时血清雌二醇质量浓度分别为(23.53±1.65)pg/L和(47.89+1.05)pg/L;血清睾酮质量浓度分别为(1.84-+0.09)ng/L和(2.47+0.12)ng/L,卵巢摘除组大鼠血清雌二醇和睾酮质量浓度均明显低于伪手术组,差异均有统计学意义(t=一35.37、一12.13,P<0.O1)。两组大鼠手术前及术后1、2、3个月sIt结果差异均无统计学意义(F:0.38,P=0.55;F=0.13,P
