5、他达拉非延缓大鼠慢性移植血管硬化病变徐自强夏鹏王瑾琚金昊杨亦荣摘要目的：探讨他达拉非对大鼠慢性移植动脉血管病变保护作用。方法：实验分3组：同系移植组．供、受者为Lewis大鼠，无处理；异系移植实验组，Lewis大鼠接受Brown．Norway(BN)大鼠腹主动脉移植；干预组他达拉非0．5mg／(kg·d)。Lewis大鼠接受BN大鼠腹主动脉移植。术后4周和8周取移植动脉．组织学观察、测量内膜和中膜厚度，Westernblot检测PKG．I、RhoA蛋白表达。收集各组血浆，ELISA检测cGMP浓度。结果：异系

6、移植实验组移植动脉呈移植物血管病表现，血管内膜显著增厚；干预组血管内膜增厚。与实验组相比显著减轻(P<0．05)；干预组血浆cGMP浓度较实验组显著增高(P<0．05)；与实验组相比，干预组PKG—I显著增高、RhoA明显降低(P<0．05)。结论他达拉非通过调控cGMP／PKG．I和RhoA通路．抑制移植物血管病发展。关键词他达拉非：移植物闭塞；移植TadalafflattenuateschronicallograftvasculopathyinratXUZi—qiang，XIAPenWANJin-ju~．
