2、andReceivedJanuary23,1996;acceptedJune5,1996Themurinecoronavirus,mousehepatitisvirus(MHV),JHMstrain,inducesabiphasicretinaldiseaseinadultBALB/cmice.Intheearlyphase,Day1toDay7,aretinalvasculitisisnotedwhichisassociatedwiththepresenceofviralproteinsandinfectiousvirus.Inthelatephase,Day10toDay140,areti
3、naldegenerationisassociatedwiththeabsenceofviralproteins,infectiousvirus,andinflammatorycells.ThepurposeofthisstudywastodetermineifviralRNApersistswithintheretinaduringtheretinaldegenerativephaseofthedisease.BALB/cmicewereinoculatedbytheintravitrealroutewith4.010TCID50/5mlofvirus.ThepresenceofviralR
4、NAwasdetectedbyinsituhybridizationwithaviralcDNAprobeandviralproteinswereidentifiedbyimmunocytochemicalstaining.Duringtheacutephaseoftheinfection,viralRNAwasfoundintheretina,RPE,ciliarybodyepithelium,andtheirisepithelium.Duringthelatephaseoftheinfection,viralRNAwasalmostexclusivelyfoundwithinthereti
5、naandRPEandnotintheanteriorsegmentoftheeye.Withintheretina,viralRNAwasdetectedintheganglioncelllayer,theinnerretina,theouterretina,andtheRPEcell.ImmunocytochemicalstainingidentifiedviralproteinwithintheretinaonlyfromDay1toDay8.Thisoculardiseasewasalsoassociatedwithapersistentsystemicinfection.Bothvi
6、ralRNAandviralproteinswereidentifiedwithintheliverduringthefirst8days.However,onlyviralRNAwasdetectedintheliverfromDay8toDay60.ThesestudiesdemonstratethatMHVestablishedanacuteinfection(Day1–8)whereinfectiousvirusandviralproteinswereidentified.Thiswasfollowedbyapersistentinfectionwithintheretinaandli
