2、December4,1976Avianinfectiousbronchitisvirus(IBV)wasradiolabeledforitsnucleicacidcompo-nentbygrowthindeembryonatedchickeneggs.PurifiedvirionsofIBVhaveabuoyantdensityinlinearsucrosegradientsof1.17-1.18g/cm3andcontainapproximately4%RNAbyweight.ThegenomicribonucleicacidofIBV
3、showsthefollowingcharacteris-tics:(11Itconsistsofonesizeclassofsingle-strandedRNAhavingamolecularweightof5.5-5.7xlo6andasedimentationcoefficientofabout48S;(2)noevidenceforsubunitstructureisapparentsincetheRNAresolvesasasinglespecieswiththesameelectro-phoreticmobilityinpol
4、yacrylamidegelsbeforeandafterheatdenaturation;(3)atleast20-30%oftheRNAmoleculesextractedfrompurifiedvirionscontainsequencesofpolyadenylicacidofapproximately4Ssize;(4)RNAobtainedfromdetergent-disruptedvirusparticlesbyphenol-chloroformextractionisinfectiousforculturesofchic
5、kembryofibroblasts,givingrisetoprogenyviruswhichislethalforembryonatedeggs.Thesefindings,togetherwiththeobservationthatIBVvirionsdonotexhibitdetectabletranscriptaseactivity,supporttheconclusionthatthegenomeofthiscoronavirusactsdirectlyasamessengerRNAineukaryoticcells.INTR
