3、tedwiththespikeproteinandtworeactedwiththemembraneprotein.Thespecificityofthemonoclonalantibodiesfortheexternalstructuralproteinswasdetectedbyimmunoprecipitationsusingradiolabelledvirus.Thereactionsofthemonoclonalantibodiesshowedthatone(S1)ofthetwoglycopoly-peptidesassociate
4、dwiththespikeproteinhasastrain-specificregioninvolvedinneutralizationandhaemagglutination,andthemembraneproteinhasantigenicdeterminantswhicharepresentonthethreestrainsofIBVtested(M41,BeaudetteandD41).Avianinfectiousbronchitisvirus(IBV),incommonwithothermembersoftheCoronaviri
5、dae,containsthreestructuralproteins:spike(S;peplomer),membrane(M)andnucleocapsid(N)(Siddelletal.,1983).ThespikeproteinofIBVcomprisestwoglycopolypeptides,S1and$2,of90K(90000mol.wt.)and84Krespectively(Cavanagh,1983a,b).PartsofbothSandtheglyco-sylatedMprotein(30K)arepresentatth
6、evirionsurface.TheNprotein(54K)isassociatedwiththeviralRNA.Severalmonoclonalantibodieshavebeenestablishedwhichreactwiththethreestructuralproteinsofanothercoronavirus,murinehepatitisvirus(Collinsetal.,1982;Flemingetal.,1983).Themajorityofmonoclonalantibodiestothespikeglycopro
7、teinneutralizedtheviruswhilstthosetotheotherstructuralproteinsdidnot,althoughsomeanti-membraneproteinmonoclonalantibodiescanneutralizethevirusifcomplementispresent.Ouraimswere(i)toestablishmonoclonalantibodiestothespikeglycoproteinofIBVanddeterminewhethertheywouldneutralizet
