2、eles,California90033SummaryNeurotropicstrainsofmousehepatitisviruses(MHV)suchasMHV-A59(A59)andMHV-4(JHMV)causeacuteandchronicencephalomyelitisanddemyelinationinsusceptiblestrainsofmiceandrats.Theyarewidelyusedasmodelsofhumandemyelinatingdiseasessuchasmultiplesclerosis(MS),inwhich
3、immunemechanismsarethoughttoparticipateinthedevelopmentoflesionsinthecentralnervoussystem(CNS).TheeffectsofMHVinfectionontargetcellDownloadedfromfunctionsintheCNSarenotwellunderstood,butA59hasbeenshowntoinducetheexpressionofMHCclassImoleculesinglialcellsafterinvivoandinvitroinfec
4、tion.ChangesinclassIexpressionininfectedcellsmaycontributetotheimmunopathogenesisofMHVinfectionintheCNS.Inthiscommunication,alargepanelofMHVstrainswastestedfortheirabilitytostimulateclassIexpressioninprimaryastrocytesinvitro.Thedatashowthatthemorehepatotropicstrains,suchasMHV-A59
5、,MHV-1,MHV-2,MHV-3,MHV-D,MHV-K,andMHV-NuU,werepotentinducersofclassIexpressioninastrocytesduringacuteinfection,measuredbyjem.rupress.orgradioimmunoassay.TheKbmoleculewaspreferentiallyexpressedoverDb.Bycontrast,JHMVandseveralviralstrainsderivedfromitdidnotstimulatetheexpressionofc
6、lassImolecules.AssaysofvirusinfectivityindicatedthattheclassI-inducingactivitydidnotcorrelatewiththeabilityoftheindividualviralstraintoreplicateinastrocytes.However,exposureofthevirusesorthesupernatantsfrominfectedastrocytestoultravioletlightabolishedtheclassI-inducingactivity,in
7、dicatingthatinfectiousvirusisrequiredforclassIexpression.ThesedataalsosuggestonMarch7,2015thatclassIexpressionwasinduceddirectlybyvirusinfection,andnotbythesecretionofasolublesubstanceintothemediumbyinfectedastrocytes.Finally,analysesofA59/JHMVrecombinantviralstrainssuggestthatcl
