2、鼠骨髓内皮细胞,通过WrightGiemsa染色计数内皮细胞集落、应用免疫荧光法观察内皮细胞表型;加入不同浓度的GMCSF,通过集落形成率、MTT法和流式细胞术检测内皮细胞生长周期,观察GMCSF对内皮细胞体外扩增的影响。结果表明:EndoM诱导的骨髓细胞可以生成内皮细胞集落,MF检测呈阳性;集落形成率检测及M
3、TT法测定均证实GMCSF对内皮细胞的增殖具有明显的促进作用;生长周期检测结果显示,GMCSF处理组的细胞进入S期的比率为9.3%,而对照组为2.1%,说明GMCSF可以通过促使内皮细胞进入S期而加快细胞的分裂,促进细胞增殖;比较第1代和第4次传代后细胞的生长曲线,两者无明显差别,说明多次传代后的细胞仍保持传代早期的增殖潜能。结论:GMCSF对内皮细胞的增殖具有促进作用,经多次扩增传代后内丿支细胞的增殖潜能无明显改变。【关键词】骨髓内皮细胞GMCSF细胞增殖EffectsofGranulocyteMacrophageColonyStimulatingFactoronGrow
