2、al,SchoolofMedicine,NanjingUniversity,Nanjing,JiangsuProvince,ChinaAbstract.Inflammationplaysanimportantroleinthepathogenesisofsecondarydamageafterspinalcordinjury(SCI).Thepresentstudyexploredtheeffectofsulforaphane(SFN),apotentanti-inflammatoryextractofcruciferousvegetable

3、s,ontheexpressionoftwoinflammatorymediators,metalloproteinase(MMP)-9andTNF-α,inamurinemodelofSCI.Murinespinalcordinjurywasinducedbytheapplicationofvascularclips(forceof10g)totheduraafterathree-levelT8-T10laminectomy.Thewet/dryweightratiowasusedtoreflectthepercentageofwaterc

4、ontentofimpairedspinalcordtissueat48hrafterSCI.ThemRNAlevelsofMMP-9weredeterminedusingthereverse-transcriptasepolymerasechainreaction(RT-PCR),andproteinlevelsofTNF-αandMMP-9weredetectedbyenzyme-linkedimmunosorbentassays(ELISA)at24hrafterSCI.Gelatinzymographywasusedtodetermi

5、neMMP-9activityofspinalcordtissueat24hrafterSCI.MicetreatedwithSFNat1hrafterSCIhadlowerexpressionandactivityofMMP-9comparedtomicewithSCI.ThedecreaseofMMP-9inmicetreatedwithSFNwasassociatedwithdecreasedlevelsofspinalcordwatercontentandTNF-α.Insummary,suforaphanedecreasesMMP-

6、9andTNF-αexpressionandvascularpermeabilitychangesfollowingspinalcordinjuryinmice.Keywords:spinalcordinjury,sulforaphane,matrixmetalloproteinase-9,TNF-αIntroductioninjuryandprotecttheneuralelementsthatinitiallysurvivethemechanicalinjury.Furthermore,evenaSpinalcordinjury(SCI)
