3、chcontributestorenalinflammationandfibrosis.Nuclearfactor-erythroid-2-relatedfactor2(Nrf2)isknowntocoordinateinductionofgenesthatencodeantioxidantenzymes.Weinvestigatedtheeffectsofoleanolicacid,aknownNrf2activator,onoxidativestress-inducedrenalinflamma

4、tionandfibrosis.Methods:Onedaybeforeunilateralureteralobstruction(UUO)performedinC57BL/6mice,oleanolicacidtreatmentwasinitiatedandwascontinueduntil3and7daysafterUUO.Renalinflammationandfibrosis,markersofoxidativestress,andchangesinNrf2expressionweresub

5、sequentlyevaluated.Results:IntheobstructedkidneysofUUOmice,oleanolicacidsignificantlyattenuatedUUO-inducedcollagendepositionandfibrosisonday7.Additionally,significantlylessinflammatorycellinfiltration,alowerratioofBaxtoBcl-2expression,andfewerapoptotic

6、cellsonTUNELstainingwereobservedintheobstructedkidneysofoleanolicacid-treatedmice.OleanolicacidincreasedtheexpressionofnuclearNrf2,hemeoxygenase-1,NAD(P)H:quinoneoxidoreductase1andheatshockprotein70,anddecreasedlipidperoxidationintheobstructedkidneyofU

7、UOmice.TherewerenochangesintheexpressionoftotalNrf2andKelch-likeECH-associatedprotein1,indicatingthatoleanolicacidenhancednucleartranslocationofNrf2.Conclusions:Theseresultssuggestthatoleanolicacidmayexertbeneficialeffectsonrenalfibrosisbyincreasingnuc

8、leartranslocationofNrf2andsubsequentlyreducingrenaloxidativestress.Backgroundthereareseveralreportsthatoxidativestressplaysasig-Tubulointerstitialfibrosisisthefinalcommonpatho-nificantroleinrenaldamageinobstructedkidneys[3-5].logicalfea
