3、hesofDNAcalledspacers,acquiredfromextrachromosomalelements.Itwasrecentlydemonstratedthatthesevariablelocicanincorporatespacersfrominfectingbacteriophagesandthenprovideimmunityagainstsubsequentbacteriophageinfectionsinasequence-specificmanner.HereweshowthattheStreptococcusthermophilusCRISPR1/C
4、assystemcanalsonaturallyacquirespacersfromaself-replicatingplasmidcontaininganantibiotic-resistancegene,leadingtoplasmidloss.Acquiredspacersthatmatchantibiotic-resistancegenesprovideanovelmeanstonaturallyselectbacteriathatcannotuptakeanddisseminatesuchgenes.WealsoprovideinvivoevidencethattheC
5、RISPR1/Cassystemspecificallycleavesplasmidandbacteriophagedouble-strandedDNAwithintheproto-spacer,atspecificsites.OurdatashowthattheCRISPR/CasimmunesystemisremarkablyadaptedtocleaveinvadingDNArapidlyandhasthepotentialforexploitationtogeneratesafermicrobialstrains.Clusteredregularlyinterspaced
