3、alth,FacultyofMedicine,UniversityofSouthampton,Southampton,UKItiswellestablishedthatgenotypeplaysanimportantroleintheageingprocess.However,recentstudieshavesuggestedthatepigeneticmechanismsmayalsoinfluencetheonsetofageing-associateddiseasesandlongevity.Epigeneticsisdefi
4、nedasprocessesthatinduceheritablechangesingeneexpressionwithoutachangeintheDNAnucleotidesequence.ThemajorepigeneticmechanismsareDNAmethylation,histonemodificationandnon-codingRNA.Suchprocessesareinvolvedintheregulationoftissue-specificgeneexpression,celldifferentiationa
5、ndgenomicimprinting.However,epigeneticdysregulationisfrequentlyseenwithageing.Relativelylittleisknownaboutthefactorsthatinitiatesuchchanges.However,thereisemergingevidencethattheearlylifeenvironment,inpar-ticularnutrition,inearlylifecaninducelong-termchangesinDNAmethy
6、lationresultinginanalteredsusceptibilitytoarangeofageing-associateddiseases.Inthisreview,wewillfocusonthechangesinDNAmethylationthatoccurduringageing;theirroleintheageingprocessandhowearlylifenutritioncanmodulateDNAmethylationandinfluencelongevity.Understandingthemecha
