1、第45卷第6期林业科学Vol145,No162009年6月SCIENTIASILVAESINICAEJun.,20093欧美杨溃疡病的病原鉴定1131,23贺伟任飞娟郭利民李永常聚普(11北京林业大学森林培育与保护教育部重点实验室北京100083;21中国林业科学研究院森林生态环境与保护研究所北京100091;31河南省濮阳市林业科学研究所濮阳457000)摘要:从河南濮阳、山东菏泽等地的欧美杨溃疡病发病林地采集病样96份,经分离、纯化、归类出10种分离物,其中镰孢属(Fusarium)真菌占优势。以离体苗干水培接种和盆栽苗接种,经致病性测定,明确分离物Mz01为致病菌株。经形态学鉴定,Mz
2、01为茄镰孢菌(Fusariumsolani)。采用真菌通用引物对分离物Mz01进行PCR扩增,对其核糖体DNA(rDNA)内转录间隔区测序,并将测序结果在GenBank中进行同源性比对分析,分子鉴定与形态学鉴定一致。茄镰孢菌作为我国欧美杨溃疡病的病原菌系首次报道。关键词:欧美杨溃疡病;病原菌;茄镰孢菌中图分类号:S76317文献标识码:A文章编号:1001-7488(2009)06-0104-006PathogenidentificationofPopulus×euramericanaCankerDisease1131,23HeWeiRenFeijuanGuoLiminLiYongChan

5、larsand10isolateswereobtained,amongwhichmostisolatesbelongedtogenusFusarium.TheisolateswereinoculatedonbothhydroponicstemandpottedtwoyearsoldseedlingsofPopulus×euramericanacv1‘74P76’for4～5daysat25℃.Re2isolationfromthelesionformedbytheinoculationwasthesameastheinocula,demonstratingthattheisolateMz01
6、wasapathogenicstrainofFusariumsolanibasedonitsmorphological.ThesequencesoftheisolatesMz01wereamplyfiedbyPCRtechniquewithuniversalprimersoffungi,andthesequenceswerefurthercomparedwiththatinGenBank.Themolecularidentificationconfirmedthesameresultsfrommorphologyidentification.ThisisthefirstreportthatF
