3、,TheNetherlandsReceived13September2005/Accepted18November2005Theimpactofenvironmentalperturbation(e.g.,nitrogenousfertilizers)onthedynamicsofmethanefluxesfromsoilsandwetlandsystemsispoorlyunderstood.Resultsoffertilizerstudiesareoftencontradictory,evenwithinsimilarecosystems.Inthepresentstudythehypot
4、hesisofwhetherthesecontradictoryresultsmaybeexplainedbythecompositionofthemethane-consumingmicrobialcommunityandhencewhethermethanotro-phicdiversityaffectsmethanefluxeswasinvestigated.Tothisend,ricefieldandforestsoilswereincubatedinmicrocosmsandsupplementedwithdifferentnitrogenousfertilizersandmethan
5、econcentrations.Bylabeling13themethanewithC,diversityandfunctioncouldbecoupledbyanalysesofphospholipid-derivedfattyacids(PLFA)extractedfromthesoilsatdifferenttimepointsduringincubation.Inbothricefieldandforestsoils,theactivityaswellasthegrowthrateofmethane-consumingbacteriawasaffecteddifferentially.
6、FortypeImethanotrophs,fertilizerapplicationstimulatedtheconsumptionofmethaneandthesubsequentgrowth,whiletypeIImethanotrophsweregenerallyinhibited.TerminalrestrictionfragmentlengthpolymorphismanalysesofthepmoAgenesupportedthePLFAresults.Multivariateanalysesofstable-isotope-probingPLFAprofilesindicate
7、dthatinforestandricefieldsoils,Methylocystis(typeII)specieswereaffectedbyfertilization.ThetypeImethanotrophsactiveinforestsoils(Methylomicrobium/Methylosarcinarelated)differedfromtheactivespeciesinricefieldso
