1、四川犬攀顿士学位论文檬浩杰摘要目的:为研究蠓强型绿色荧光蛋白(enhancedgreenfluorescentprotein,EGFP)罄因在活纲溉中{搴为报告基因帮筛选标记酶可行稳,特构建其真核表达载体pCDNA3.1(+)-EGFP,并将它转染至Hela缨胞和大鼠骨髓闻充质于细胞(mesenchymalstemcell,MSC)中,躐察箕在这两释细胞中酌表达情况。方法:以BamHI和Not1双酶切分别从真核表达载体pCDNA3.1(+)_荦班EGFP载体pEGFP一1中切敬线性化pCDNA3。l(+)载体和晷鲍EGFP基闲片段,然磁
2、将两者连接起来,献两获得重组质糍pCDNA3.1(+)一EGFP。并分别以BamHI单酶切和BamHI、NotI双酶切进行鉴寇。以腮质体转染试剂LipofectAMINE2000分别将构建姆约重组质靛转染歪培养豹Hela细胞和大鬣雷髓闻充艨干细胞中,分别经G418筛选两周和四周厝获得EGFP阳性克隆,进行细胞爬片质放置在荧光晟微镜下鼹察。缝纂:成功构建了含EGFP基困片段的凑核表运载体pCDNA3.1(+)一EGFP,并顺利转染至Hela细胞和大鼠骨髓间充质干细胞中,EGFP疆性克隆在荧光照微镜下均嫩绿色。绪论:EGFP基因能以真核寝达
