1、河北大学硕士学位论文毛细管电泳测定虾肌肉中腺苷酸方法研究姓名:王冬梅申请学位级别:硕士专业:水生生物学指导教师:王安利;王维娜2000.6.1摘要摘要毛细管电泳(CE)是当今最有效的分离技术之一.自八十年代初得到了迅猛发展。毛细管区带电泳(CZE)和胶束电动毛细管色谱(idECC)已经用于核苷酸分析,综述了毛细管电泳法测定核苷酸的影响因素和应用实例,讨论了毛细管电泳法测定核苷酸的优势及存在的问题。建立了咬柬电动毛细管色谱测定腺营酸的方法。分析使用T卜2型高效毛细管电泳仪、48cm(有效长度37.5C91)×7SlamI.D.毛细管,254nm在柱紫外检测。首先在表面活性剂的浓
2、度、电泳缓冲液的pH和电压方面进行了优化,最佳分离条件是:50mmol/L磷酸氢二钠(Na2HPO{)+20mmol/L十六烷基三甲基溴化铵(CRAB)+lmmol/L乙二铵四乙酸二钠(EDTANa,)的电泳缓冲液(pH8.O),一15KV的电压。三种腺苷酸在10min内得到高效分离,分离度大于3。此方法用于虾肌肉中的腺苷酸测定.结果证明MECC为组织中的腺苷酸分析提供了一个简单、快速、灵敏、可靠的方法。/,、、把上述~tECC测定腺菅酸的方法应用于日本沼虾蜕壳周期肌肉中能量变化的研究。ATP浓度、TAN、AEc和ATP/ADP变化呈s一型,正蜕壳时最低,蜕壳后24小时时最高
