7、,实验分七组(n=6):①正常组:修正洛氏液连续灌流110min;②模型组:修正洛氏液稳定灌流20min后停灌30min,复灌修正洛氏液60min;③GLPP给药组:修正洛氏液稳定灌流20min后停灌30min,复灌液中分别加入不同浓度的GLPP(6.25、12.5、25、-1-150、100mg·L)灌流60min。实验过程实时监测心率(HR,min)、心脏-1张力(Tmin,g)、收缩幅度(△T,g)、最大收缩速度(+dT/dtmax,g·s)、最-1-1大舒张速度(-dT/dtmax,g·s)、冠脉流量
8、(CF,d·min)反应心功能。实验结束,留取心室肌光镜观察心肌组织学结构、电镜观察心肌细胞超微结构,TUNEL检测心肌细胞凋亡,Westernblot检测心肌细胞Caspase-3的活性。结果:1心搏曲线在心脏缺血再灌注期间,模型组收缩力减弱,心率减慢,出现心律失常;给药组收缩力增强,心率增快,节律整齐。2各项参数2.1心肌张力模型组在缺血20min后张力增加,对照组和模型组在缺血20min时的张
