2、d.MethodsForty-nine3-month—oldNewZealandWhiterabbitswereincluded,weighing2-2.5Kg,48femaleand1male.BMSCsoftherabbitswereisolatedbydensitygradientseparationmethod,thethirdpassagecellsweremarkedbyDiI.TheanimalmodelofONFHwereestablishedwithfortyeightfemalerabbitsbyinjectingliquidnitrogen,andfemoralhead
3、wasnotdislocated,whichweredividedinto3groups,16rabbitsineachgroup,groupAonlyestablishedanimalmodelascontrol,autologousBMSCsmarkedbyDilwastransplantedintheONFHmodelsofthegroupB.AllogenicBMSCsmarkedbyDilwastransplantedinONFHmodelsofthegroupC.ThefemoralheadwereobservedbyX—ray,HEstainingandMassontrichr
4、omestainingandtheregeneratingtrabecularvolumepercentagesweredeterminedat2,4,6and8weeksafteroperationrespectively.Theexamplesoftheheart,lung,liver,spleenandkidneywereobtained.ThetransplantedBMSCsweretracedbyfluorescencemicroscope,theSRYgeneexpressionwasdetectedbypolymerasechainreaction(PCR)forcellss
5、urvival.ResultsTheX·rayshowednecrosisinthefemoralheadofgroupAgradually.HEandMassontrichromestainingresultsshowedthatcomparedwiththegroupA,therecoveryconditionofthenecroticfemoralheadinthegroupBandCWasbetter.Ateachtime·pointofgroupBandC,theregeneratingtrabecularvolumepercentageswerehigherthanthatoft
6、hegroupAsignificantly.TheresultbetweengroupBandCWasinconspicuous.CellsmarkedbyDilwerenotbeenfoundedinthetissueoftheheart,lung,liver,spleenandkidneyateachtimepoint.PCRshowedthattheexpressionofSRYgeneWasnotobservedattheheart,lung,liver,spleenandkidneyofthreegroupsateachtimepoint.TheexpressionofSRYgen
