1、摘要蓖麻是内蒙地区非常具有区域特色的世界十大油料作物之一。蓖麻油由脂肪酸组成,主要成分是蓖麻油酸。磷脂酶C在控制蓖麻油酸生物合成过程中起关键性的作用,它催化2一油酰基卵磷脂形成卜酰基一2一油酰基甘油,使非羟基脂肪酸进入三酰基甘油。通过基因工程手段设计并构建含有磷脂酶C基因的RNAi表达载体,以期在植物体内转录出含有部分磷脂酶C基因发夹结构的dsRNA,发挥干扰磷脂酶C基因表达的作用,从而提高总蓖麻油含量。本试验以通蓖5号蓖麻为研究对象,提取其发育阶段的种子总RNA,逆转录成cDNA,通过降落PCR得到长度分别为803bp和45
2、8bp的磷脂酶C基因片段C1和C2,采用Sanger双脱氧链终止法对Cl和C2进行测序,测序结果与Genbank中蓖麻磷脂酶C基因的mRNA进行同源性分析,同源性分别达到99.88%和99.13%,成功克隆了蓖麻磷脂酶C基因。并采用一步法成功构建了蓖麻磷脂酶C基因的RNAi表达载体,为通过RNAi干扰技术获得总蓖麻油含量高的蓖麻新材料奠定了基础。关键词:蓖麻;磷脂酶C;RNAi;表达载体CloningofAPhospholipaseCandConstructionoftheRNAiExpresionVectorinCastor
