1、温州医学院硕士学位论文糠尿病大鼠眼组织中醛脱氢酶活性及表达量的实验研究姓名:王明玲申请学位级别:硕士专业:眼科学指导教师:肖天林2012-05-21温州医学院硕士学位论文糖尿病大鼠眼组织中醛脱氢酶活性及表达量的实验研究中文摘要背景以往的研究发现糖尿病并发症的发生与脂质过氧化有关,而醛脱氢酶是已知的抗脂质过氧化物酶之一,目前醛脱氢酶在糖尿病眼组织中的活性与表达量变化的研究尚不多见。目的研究不同时期糖尿病大鼠眼组织中醛脱氢酶的活性与含量,探讨该酶在糖尿病所致眼部疾病的变化及作用。方法清洁级170—1809左右的SD雄性大鼠28只按随机数字表法分为糖
2、尿病组与正常对照组;糖尿病组腹腔注射65mg/kg溶于柠檬酸钠缓冲液的链脲佐菌素(streptozotocin,STZ),正常对照组腹腔注射等量柠檬酸钠缓冲液。于造模成功后的2月与4月两个时间点处死各组7只大鼠分别制备大鼠眼角膜、晶状体及视网膜匀浆液,测定蛋白质浓度。采用M5多功能酶标仪测量角膜、晶状体及视网膜各组织中醛脱氢酶(ALDH)活性;酶联免疫吸附(ELISA)法检测醛脱氢酶的表达量。结果造模成功后2月与4月时糖尿病组大鼠眼角膜、晶状体及视网膜中ALDH活性及含量均明显高于相应时间点的正常对照组,差异有统计学意义(P<0.001)。4月
