1、■t曰面feE文目的:研究附子与半夏不同炮制品配伍前后乌头类生物碱成分的含量变化及种类增减,探讨附子与半夏配伍相反理论的物质基础。方法:根据临床附子的使用习惯,选取不同产地生附子2种、常用附子炮制品7种、生半夏1种及半夏炮制品3种作为研究对象。采用HPLC-MSn法及ESI-MSn法对附子单煎以及附子半夏不同配伍组合、不同煎煮条件下煎煮液中乌头类生物碱的含量和种类进行比较。结果:1.生附子单煎60min较单煮lOmin,其煎液中11种生物碱含量有明显降低。2.生附子同1倍量生半夏共煎液较生附子单煎液,双酯型生物碱含量有明显增加;生附子同2倍量生半夏共煎液较生附子同1倍量生半夏共煎液,双
2、酯型生物碱含量有明显增加。3.附子分别同生半夏、清半夏、姜半夏共煎后,双酯型生物碱含量有明显增加;附子同法半夏共煎后双酯型生物碱含量低于附子单煎液。结论:1.适当延长煎煮时间能降低附子水煎液中的双酯型生物碱含量。2.生半夏、清半夏、姜半夏有抑制附子水煎液中双酯型生物碱分解的作用,而法半夏没有上述作用。3.生半夏用量的增加在一定范围内提高了对附子水煎液中双酯型生物碱分解的抑制能力。关键词附子;半夏;HPLC-MSn;ESI-MSn;双酯型生物碱AnalysisofAlkaloidsintheCombinationofRadixAeonitiLateralisPreparatawithDi
