1、东南大学硕士学位论文异丙酚对大鼠肺缺血再灌注损伤保护作用的实验研究姓名:张桂武申请学位级别:硕士专业:临床医学指导教师:刘志勇2012-06-01中义摘要异丙酚对大鼠肺缺血再灌注损伤保护作用的实验研究研究生:张桂武导师:刘志勇东南大学医学院[摘要]目的:通过建立大鼠肺缺血再灌注模型,观察异丙酚(Propof01)对大鼠肺缺血再灌注损伤(IschemiaReperfusionInjury,IRI)的保护作用,并探讨其作用机制。方法:随机将30只雄性SD大鼠分为3组(每组10只):假手术组(A)(左侧开胸后不阻断肺门);肺缺血再灌注组(B)(左侧肺门阻断45min再灌注120m
2、in),异丙酚组(C)(左肺门阻断前30min时给予股静脉注射异丙酚5mg/kg,继用微量泵以50mg/kg/h的速度持续静脉输注,左侧肺门阻断45min再灌注120min)。三组在开胸前均给予气管插管,机械通气。再灌注结束时抽取左心室血检测动脉血氧分压(Pa吼),获取左肺组织,光镜下观察左肺组织和细胞损伤,计算左肺湿干重比,评价肺水肿程度,制作肺组织匀浆并检测其中丙二醛(Matondialdehyde,MDA)含量、肿瘤坏死因子一Q(TumornecrosiSfactor—a,TNF—Q)、白细胞介素一6(interleukin一6;IL-6)和超氧化物歧化酶(super
