1、东南大学硕士学位论文SDF--1在正常及退变新西兰兔椎间盘中的表达分布姓名:庄宇申请学位级别:硕士专业:临床医学指导教师:吴小涛2012-05-25中文摘要SDF一1在正常及退变新西兰兔椎间盘中的表达分布东南大学临床医学院研究生:庄宇导师:吴小涛教授目的:探明兔退变椎间盘内SDF一1的表达分布梯度,为利用SDF-1/CXCR4趋化信号轴调控干细胞在退变椎间盘内的迁移分布行为提供理论基础。方法:取成年新西兰大白兔50只,随机分为退变诱导组(40只)与正常对照组(10只)。退变组通过纤维环穿刺抽吸髓核法诱导腰3-
2、6三个节段退变,正常组单纯行腰3—6节段暴露。分别于术后第4周、12周处死正常组实验兔5只和退变组实验兔20只,获取腰3—6三节段椎I、日J盘,组织切片后HE染色、免疫组织化学法检测II型胶原、MMP—13,观察椎问盘组织学退变程度,免疫组织化学法检测纤维环、髓核、软骨终板区域SDF一1表达,与正常组比较,观察SDF-1在椎间盘内的表达分布梯度。结果:l、造模术后兔椎问盘组织内的二型胶原表达明显低于正常对照组,而MMP一13的表达明显高于正常对照组,P3、高于正常对照组,且术后第12周的表达高于术后第4周,差异有统计学显著性意义(p<0.05)。术后第4周和第12周,SDF-I在髓核组织内的表达显著高于纤维化与软骨终板区域,形成退变椎间盘内SDF一1的表达分布梯度。结论:SDF-1在兔椎间盘的纤维化、髓核、软骨终板区域表达。退变椎间盘上调SDF-I的表达,髓核组织内的表达量显著高于纤维化与软骨终板区域,可形成退变椎问盘内SDF-I的表达分布梯度。[关键词]基质细胞源性生长因子-I(SDF一1);兔椎问盘退变模型:浓度梯度;免疫组织化学英文摘要■_■■■●·一I4、11eexpressionandSIgnlticanceofSDF-1innormalanddegeneratedintervertebraldiscsofNewZealandwhiterabbitsPostgraduate:ZhuangYuSupervisor:Pro:WuXiaotaoMedicalCollegeofSoutheastUniversityEnglishAbstractObjective:ThisstudyaimstofindoutwhetherthereisaformationofSDF.5、1concentrationgradientinthedegenemtedintervertebraldiscsofrabbitstoprovideatheoreticalbasisfortheSDF-1/CXCR4chemokinesignalingaxisregulationofstemcellmigrationanddistributionwithinthedegeneratedintervertebraIdiscs.Methods:Selected50adultNewZealandwhiterabbi6、ts,whichweremndomlydividedintodegenerationgroup(n=40)andnormalcontrolgroup(n=l0).DegenerationgroupwereinducedL3/4,L4/5andL5/6lumbarintervertebraldiscsdegenemtionbyaspimtionthroughtheannulusfibrosusandnucleuspulposus.NormalgroupweretreatedwithL3/4,L4/5andL5/7、6exposed.4weeksaftersurgery,getL3/4,L4/5andL5/6lumbarintervertebraldiscsofnormalcontrolgroupand20ofdegenerationgroup,then12weeksaftersurgery,getthelumbarintervertebraldiscsoftherest20ofdegenerationgroup.TissuesectionswithHEstaining,immunohistochemistrywasus8、edtotesttheexpressionoftheTypeIIcollagenandMMP-13toobservethedischistologicaldegeneration.ImmunohistochemistrywasagainusedtotesttheexpressionofSDF一1withinannulus,nucleuspulposus,cartilageendplateregion
