1、陕西中医学院硕士学位论文醒脑解郁胶囊对脑卒中后抑郁模型大鼠海马突触素SYN的影响姓名:杨文佳申请学位级别:硕士专业:中西医结合临床指导教师:闫咏梅201204摘要实验目的:通过观察醒脑解郁胶囊对脑卒中后抑郁(Post—strokedepression,PSD)大鼠治疗前后体重及行为学、大鼠海马SYNmRNA的表达的影响,探讨和揭示醒脑解郁胶囊在中医基础理论指导下,治疗PSD的靶点及其对海马神经元的保护作用,为临床治疗提供相应的实验依据和临床参考。实验方法:将SD大鼠共58只随机分为5组:空白对照组组、卒中后抑郁模型对照组、氟西汀组、醒脑解郁胶囊大剂量组、醒脑解郁胶囊小剂
2、量组。应用大脑中动脉闭塞法(MCAO)联合慢性的不可预见的温和应激刺激(CMS)、孤养法建立病症结合动物模型:脑卒中后抑郁(PSD)大鼠模型。分别测评实验各组大鼠的体重、蔗糖水消耗量、敞箱试验得分,观察醒脑解郁胶囊对PSD大鼠以上各项指标的影响。采用免疫组化法和RT-PCR法检测和观察大鼠海马区SYNmRNA表达的影响。实验结果:1.观测醒脑解郁胶囊对PSD大鼠体重即糖水消耗量的影响:与空白对照组相比,PSD模型对照组的大鼠体重下降明显,蔗糖水消耗量降低锐减,醒脑解郁胶囊大剂量组、醒脑解郁胶囊小剂量组与氟西汀组大鼠体重、蔗糖水消耗量也不同程度增加。2.观测醒脑解郁胶囊对
