1、徐州医学院硕士学位论文茶多酚对人卵巢癌细胞株SKOV3的顺铂增敏作用及机制姓名:王倩申请学位级别:硕士专业:妇产科学指导教师:陆晓媛;张蓓2012-03徐州医学院硕士学位论文茶多酚对人卵巢癌细胞株SKOV3的顺铂增敏作用及机制中文摘要目的探讨茶多酚(11P)对人卵巢癌细胞株SKoV3增殖及顺铂(DDP)敏感性的影响及机制。方法MTT法检测单独使用不同浓度TP、DDP及低毒剂量TP联合DDP对人卵巢癌细胞株SKOV3的增殖抑制作用,分别计算抑制率、半数抑制浓度(IC50)及增敏倍数,DAPI核染色法荧光显微镜下观察细胞凋亡形态
2、变化,ArulexinV-FITC双染流式细胞术分析细胞凋亡,W.estenlB10t方法检测细胞中Al(t及p.Al(t的表达。结果TP、DDP单药均以剂量依赖的方式抑制人卵巢癌细胞株SKOV3的增殖。低毒剂量的TP(6.25m∥L)联合不同浓度的DDP作用于细胞后,增殖抑制率均明显增加,IC50由单用DDP时的6.555m∥L下降到3.021m∥L,敏感性提高到单用DDP的2.17倍。其中,6.25m∥LTP、2.5m班DDP和6.25m∥LTP联合2。5m∥LDDP组对细胞的增殖抑制率分别为(11.47±2.07)%、
