1、遵义医学院硕士学何论文中文摘要糖皮质激素对哮喘大鼠Ai0lOS基因表达及血清IgE的影响中文摘要目的:观察哮喘模型大鼠外周血淋巴细胞AiolosmRNA的表达和血清免疫球蛋白I班的量,检测地塞米松(DXM)干预前后AiolosmRNA及血清IgEi拘变化,分析Aiolos基因与免疫球蛋白IgE在哮喘发病中的关系,探讨Aiolos基因调控在哮喘发病和糖皮质激素治疗哮喘中的机制,为进一步探讨哮喘的发病机制打下基础。方法:将30只SD大鼠随机分为正常对照组、哮喘模型组和地塞米松组,每组lO只。哮喘模型组以卵清蛋白(OVA)致敏和激发复制哮喘模型,首日致敏采
2、用腹腔内注射含l%卵清蛋白和10%氢氧化铝的生理盐水混悬液lml,第8天相同剂量、相同方法加强致敏1次,从实验第15天开始,雾化吸入1%卵清蛋白溶液激发,每日1次,每次30分钟,连续2周。地塞米松组分别于每次雾化前ld,时地塞米松腹腔注射干预。正常对照组以生理盐水代替卵清蛋白致敏和激发。末次激发24dx时后,下腔静脉取血,开胸取肺组织,肺组织切片HE染色,再结合大鼠激发后反应来鉴定模型是否已建立。RT.PCR法检测外周血淋巴细胞Aiolos基因的表达,ELISA法测定血清IgE的量。结果:成功建立哮喘大鼠模型。与正常对照组相比,哮喘模型组大鼠出现了明
