1、人皮肤鳞状细胞癌体外三维模型的建立及高侵袭性鳞状细胞癌的生物学评价杨晓鲲,杨亚东,杨桂红,唐书谦,伍津津*(第三军医大学大坪医院野战外科研究所皮肤科,重庆,400042)摘要目的:建立人皮肤鳞癌细胞体外三维培养模型,分离高侵袭性细胞亚群,并比较其生物学特性。方法:利用复方壳多糖组织工程皮肤,成功建立人皮肤鳞癌细胞体外三维培养模型,消化分离得到高侵袭性鳞癌细胞亚群,比较肉桂醛和干扰素作用后对不同侵袭特性细胞亚群的增殖抑制效果不同,同时比较不同侵袭特性细胞亚群在裸鼠中成瘤效果的不同。结果:我们成功地建立了人皮肤鳞状细胞癌体外三维培养模型,经过肉桂醛和干扰素对
2、皮肤鳞癌细胞的增殖抑制实验我们发现,随着药物浓度的增加,鳞癌细胞被抑制率也随之增高,具有统计学意义(P<0.01)。其中肉桂醛在浓度为400umol/L、作用时间为72h时,对2组细胞的抑制率分别达到85.65±4.31%和73.45±3.55%,而干扰素在浓度为400umol/L、作用时间为72h时,对2组细胞的抑制率则分别达到了51.65±2.37%和45.35±6.26%,同时这2组实验分组细胞之间比较,相同浓度药物和相同药物作用时间对肿瘤细胞的增殖抑制也有统计学意义(P<0.01),另外,两种不同侵袭特性的细胞亚群的成瘤特性同样有统计学意义(P<
