1、KLF4对胃癌SGC-7901细胞株体外生长增殖的影响黄镇，王子卫，张能，查郎，吴钊（400016重庆，重庆医科大学附属第一医院胃肠外科）［摘要］目的探讨Krüppel样因子4（krüppel-likefactor4，KLF4）对人胃癌SGC-7901细胞株生长增殖的影响。方法以人胃癌SGC-7901细胞株为研究对象，脂质体介导转染pcDNA3.1IE-KLF4重组质粒并建立KLF4稳定过表达细胞株，以未转染KLF4基因和转染空质粒载体的胃癌细胞株为对照。用RT-PCR和免疫细胞化学法检测KLF4mRNA和蛋白的表达，流式

2、细胞术检测细胞周期及增值指数。结果RT-PCR和免疫细胞化学法检测示转染组细胞与未转染组和空载体组相比较，KLF4mRNA和蛋白均明显表达（P＜0.05）。流式细胞术检测显示转染组G1期细胞增多［(65.08±3.43)﹪，P＜0.05］，而S期和G2/M期细胞减少［(30.39±1.80)﹪，(4.53±1.66)﹪，P＜0.05］，增殖指数亦降低［(34.92±3.42)﹪，P＜0.05］。结论KLF4对SGC-7901细胞株体外生长增殖具有抑制作用。［关键词］Krüppel样因子4；胃癌；转染；细胞周期；增殖［中图法
