3、mAe.biuncialisintowheatviascreeningitsadditionlines,substitutionlines,ortranslocationlines.Inthestudy,Ae.biuncialiswascrossedtowheatlineCN19toproduceF1hybridplants.andthenthehybridplantsweretreatedwithcolchicine,weobtainedwheat-Ae.Biuncialisamphiploid(AABBDDUUMM,2n=10x=70).Afte
4、rpreliminaryidentificationofthedescendantsofthehybridizationbetweenCN19andwheat-Ae.biuncialisamphiploid,weobtainageneticallystableline12-5-2,whichcontainsspecialhighmolecularweightgluteninsubunitsfromAe.biuncialis.Chromosomenumbersinroot-tipcellsof12-5-2were2n=44,anditwasimmune
5、tobothstriperustandpowderymildew.Itwasconfirmedthat12-5-2wasa1Ubdisomicadditionline(ADL1Ub)byfluorescenceinsituhybridizationandC-banding.seedproteinSDS-PAGEanalysisshowedthat12-5-2notonlyownedallhighmolecularweightgluteninsubunits(7+8and2+12)fromitsparentCN19,butalsohad2newsubu
