4、41657TandG4234CSNPsinthenonsmallcelllungcancerpatientsthoseofthecontrols(allP0.05).gCl21.5mmol/L(C41657T)或1.25mmol/L(G4234C),TaqDNA聚合酶2U,dNTPs160μmmol/L和上、下游引物各200nmol/L.PCR反应条件为:94℃5min预变性后,94℃变性45s,72℃(C41657T)或70.1℃(G4234C)退火45s,72℃延伸1min,35个循环后,72℃继续延伸7min.XRCC2C41657TPCR产物经限制性内切酶Mva
5、Ⅰ166.7nkat于37℃酶切16h后进行40g/L脂糖凝胶电泳.C/C基因型为315bp,42bp和59bp3个DNA片段,T/T基因型为357bp和59bp两个DNA片段,C/T基因型为315bp,357bp,42bp和59bp4个DNA片段(图1).XRCC2G4234CPCR产物经限制性内切酶MluⅠ166.7nkat于37℃酶切20h后,进行40g/L琼脂糖凝胶电泳.G/G基因型为251bp一个DNA片段,G/C基因型为251bp,230bp和21bp3个DNA片段,C/C基因型为21bp和230bp两个DNA片段(图2).为进行基因型检测的质量控制,每一批
