1、第四军医大学硕士学位论文在腹腔内注射后约40min,并且持续到注药后70min仍未完全消失。而健侧、拉科酰胺低剂量组及溶剂对照组PWMT值于加药前后均没有明显改变(P>0.05)。实验二:拉科酰胺对大鼠伤害性感觉通路突触传递的作用目的:观察拉科酰胺对大鼠脊髓伤害性细纤维(Aδ纤维、C纤维)介导的兴奋性突触后电流(eEPSCs)的影响。方法:雄性SD大鼠,3~5周龄,80-100g,人工脑脊液心脏灌注后分离脊髓,在脊髓腰膨大做带后根的矢状位切片。采用全细胞记录的方式记录脊髓背角Ⅱ层神经元,输入阻抗<10MΩ,电压钳制-70mv
2、,记录稳定10min后,观察拉科酰胺对伤害性细纤维支配的SDH神经元突触传递的影响。结果:(1)拉科酰胺能够抑制伤害性细纤维介导的单突触II层神经元恒压电刺激后根诱发的eEPSCs峰值,其作用呈现剂量依赖性。(2)拉科酰胺并不影响脊髓背角II层神经元sEPSCs放电幅度,但对频率表现为抑制(P<0.01;K-Stest)。结论:1.拉科酰胺后对伤害性细纤维(Aδ纤维、C纤维)eEPSCs表现出抑制作用,且呈剂量依赖性。2.拉科酰胺对脊髓背角浅层神经元sEPSCs的频率表现为抑制作用,但不影响其放电幅度,提示拉科酰胺的作用位点
