2、ndBALB/cmiceirradiatedwith60Coγ-ray,inordertoelucidatetheradioprotectionmechanismofSelaginellatamariscina.Methods:1.WeutilizetheCCK-8AssayandGimsastainingtoobservecellproliferationandcellclonogenicsurvivalofV79cellsirradiatedwith1-8Gy60COγ-ray.V79cellsarestainedwithHoec
3、hst33258andtrypanbluetoobserveapoptosisanddeathrateinducedby4,8Gyγ-irradiation.V79cellsaredetectedwithReactiveOxygenSpeciesAssayKitandPIsimplestainingtoobserveROSlevelandcellcycle.V79cellmodelisbuilttoquickscreenoutanti-radiationdrugsinvitrofromcompoundsofSelaginellaTam
4、ariscinainthisexperiment.2.Carriesonthepreliminaryscreeningofanti-radiationeffectivemonosaccharidesconstituentsfromSelaginellaTamariscinathroughtheV79cellmodel,xyloseofthemshowradioprotectiveactivitiesinvivothroughthemousemodelirradiatedby6Gy.3.Screenoutofanti-radiation
5、activenessfromSelaginellaTamariscinadifferentextractingpartsthroughAHH-1cellmodelirradiatedby6Gy,andeffectivepartsofthemareverificatedthroughthemousemodelirradiatedby8Gy.Thenanalyzethechemicalconstituentof80%alcoholpartbyHPLC.Toevaluatetheradioprotectiveeffectofamentofl
6、avonethroughtheV79cellmodelandthemousemodelirradiatedby6Gy,thecellviability,cellclonogenicsurvival,cellcycleand30-daysurvivalratearedetected.Results:1.WefindthatV79celldeathrateincreasewith60COγ-raydoes-dependentmanners.Alongwiththeincreaseofradiationtimeordose,thecellv
