1、神经病理性疼痛大鼠行为学及生长相关蛋白表达的变化厉彩霞林海王均炉△温州医学院附属第一医院麻醉科[摘要]目的:观察大鼠坐骨神经结扎后神经病理性疼痛行为学以及生长相关蛋白-43(GAP-43)表达的变化。方法:采用坐骨神经结扎损伤(CCI)模型,75只SD大鼠,随机分为三组(n=25)。对照组(con组):不接受任何手术操作;假手术组(sham组):只分离坐骨神经,不予以结扎;CCI组:结扎坐骨神经。于术前2d,术后1、3、5、7、10、14、21、28d测定各组大鼠热缩足反射潜伏期(TWL),机械缩足反应阈值(MWT)
2、,并于术后3、7、14、21、28天各组随机抽取5只大鼠杀死,采用免疫组织化学方法测定脊髓背根节GAP-43表达的变化。结果:与sham组和比较,CCI组术后各日TWL,MWT明显降低(P<0.01或<0.05),sham组和con组行为学无显著差异(P>0.05)。CCI组较其它组GAP-43表达增高,且此蛋白在CCI组中术后第三天表达开始增高,第七至十四天达最高峰,以后逐渐下降。结论:CCI造模成功后大鼠痛觉及机械痛觉均过敏,且GAP-43表达增高。[关键词]神经病理性疼痛行为学改变GAP-43Behaviorl
