1、DEN联合NMOR诱导建立具有转移潜能的肝癌模型作者:谢胜学许戈良荚卫东王志华李建生马金良葛勇胜【摘要】目的探讨二乙基亚硝胺(diethylnitrosamine,DEN)联合N亚硝基吗啉(Nnitrosomorpholine,NMOR)诱导建立具有转移潜能的SD大鼠肝癌模型以及合适的剂量和周期。方法5周龄雄性SD大鼠110只,分成4组:DEN+NMOR组(60只)、DEN组(20只)、NMOR组(20只)以及正常对照组(10只)。第1天DEN+NMOR组和DEN组大鼠按100mg/kg腹腔注射DEN1次,NMOR组和对照组给予等量的生理盐水腹腔注射。第2天DE
2、N+NMOR组和NMOR组给予含质量分数为1×10-4NMOR的饮水自由饮用,DEN组和对照组饮用灭菌自来水。实验共进行20周。结果16周时DEN+NMOR组、DEN组、NMOR组及正常对照组肝癌发生率分别为60%(6/10)、20%(1/5)、0(0/5)、0(0/2);20周时分别为88%(21/24)、17%(1/6)、14%(1/7)、0(0/2)。DEN+NMOR组16周及20周肝癌肺转移率分别为33%(2/6)和48%(10/21),其他组未发现肺转移。结论(1)用DEN启动,NMOR促进的方法诱导建立SD大鼠肝癌模型操作简便、效果确切、成瘤周期短,有一
