2、发2型糖尿病模型,成模大鼠再随机分成非药物干预糖尿病组(DM组,”=15)和.硫辛酸干预组(ALA组,60mg/kg,=15),干预8周。干预结束后留取血清和肝脏组织,行生化及氧化应激指标检测,组织形态学检测,并应用Westernblot的方法测定大鼠肝脏组织脂代谢相关蛋白固醇调节元件结合蛋白.1(sterolregulatoryelementbindingprotein.1,SREBP.1)的表达。结果造模组大鼠血清TG【(1.6±0.69)mmol/L]和TC[(3.14±1.1)mmol/L]含量明显升高。A治疗后,血清T
3、G[(0.69±0.13)mmol/L]和TC[(1.67±1.0)mmol/L]明显降低,肝功能和氧化应激得到了改善,肝脏中的脂质沉积明显减少。SREBP.I的蛋白表达水平明显减低。结论ALA干预可改善高脂饮食联合STZ诱导的SD大鼠的肝脏脂质沉积,其机制可能与改善氧化应激及减少SREBP.1的表达有关。【关键词】硫辛酸;糖尿病,2型;肝脏脂质沉积a-lipoicacidrelievedhepaticsteatosisbyinhibitedHpogenesisindiabetesmellitusratSHIZhi-zhou,G
